Design: The nifH gene (~359-bp), which used as a functional gene indicator of both diazotroph diversity and activity was amplified from environmental DNA extracts using a nested PCR protocol. Two sets of degenerate primers were used to target the region: for the 1st round PRC nifH3 reverse primer (5'-ATRTTRTTNGCNGCRTA-3') and nifH4 forward primer (5'-TTYTAYGGNAARGGNGG -3') are used. Then the 2nd round PCR is conducted with nifH1 forward primer (5'-TGYGAYCCNAARGCNGA-3') and nifH2 reverse primer (5'-ADNGCCATCATYTCNCC-3') (J. Zehr and Turner (2001); J. P. Zehr and McReynolds (1989).
